Did bacteria spark evolution of multicellular life?

ScienceDaily (Oct. 23, 2012) ? Bacteria have a bad rap as agents of disease, but scientists are increasingly discovering their many benefits, such as maintaining a healthy gut.

A new study now suggests that bacteria may also have helped kick off one of the key events in evolution: the leap from one-celled organisms to many-celled organisms, a development that eventually led to all animals, including humans.

Published this month in the inaugural edition of the new online journal eLife, the study by University of California, Berkeley, and Harvard Medical School scientists involves choanoflagellates (aka "choanos"), the closest living relatives of animals. These microscopic, one-celled organisms sport a long tail or flagellum, tentacles for grabbing food and are members of the ocean's plankton community. As our closest living relative, choanos offer critical insights into the biology of their last common ancestor with animals, a unicellular or colonial organism that lived and died over 650 million years ago.

"Choanoflagellates evolved not long before the origin of animals and may help reveal how animals first evolved," said senior author Nicole King, UC Berkeley associate professor of molecular and cell biology.

Since first starting to study choanoflagellates as a post-doc, King has been trying to figure out why some choanoflagellates live their lives as single cells, while others form colonies. After years of dead ends, King and undergraduate researcher Richard Zuzow discovered accidentally that a previously unknown species of bacteria stimulates one choanoflagellate, Salpingoeca rosetta, to form colonies. Because bacteria were abundant in the oceans when animals first evolved, the finding that bacteria influence choano colony formation means it is plausible that bacteria also helped to stimulate multicellularity in the ancestors of animals.

"I would be surprised if bacteria did not influence animal origins, since most animals rely on signals from bacteria for some part of their biology," King said. "The interaction between bacteria and choanos that we discovered is interesting for evolutionary reasons, for understanding how bacteria interact with other organisms in the oceans, and potentially for discovering mechanisms by which our commensal bacteria are signaling to us."

No one is sure why choanoflagellates form colonies, said one of the study's lead authors, UC Berkeley post-doctoral fellow Rosanna Alegado. It may be an effective way of exploiting an abundant food source: instead of individual choanoflagellates rocketing around in search of bacteria to eat, they can form an efficient bacteria-eating "Death Star" that sits in the middle of its food source and chows down.

Whatever the reasons, colonies of unicellular organisms may have led the way to more permanent multicellular conglomerations, and eventually organisms composed of different cell types specialized for specific functions.

Sequencing the choanoflagellate genome

King's 12-year search for the trigger of choanoflagellate colony development was reignited in 2005 when she started to prime cultures of the choanoflagellate S. rosetta for a genome sequencing project. The sequencing of another choanoflagellate, the one-celled Monosiga brevicollis, gave some clues into animal origins, but she needed to compare its genome to that of a colony-forming choanoflagellate.

Surprisingly, when Zuzow tried to isolate the colony-forming choanoflagellate by adding antibiotics to the culture dish to kill off residual bacteria, strange things happened, said King.

"When he treated the culture with one cocktail of antibiotics, he saw a bloom of rosette colony formation," she said, referring to the rose petal-shaped colonies that were floating in the culture media. "When he treated with a different cocktail of antibiotics, that got rid of colony formation altogether."

That "rather mundane but serendipitous observation" led Zuzow and Alegado to investigate further and discover that only one specific bacterial species in the culture was stimulating colony formation. When other bacteria outnumbered it, or when antibiotics wiped it out, colony formation stopped. Alegado identified the colony-inducing bacteria as the new species, Algoriphagus machipongonensis. While she found that other bacteria in the Algoriphagus genus can also stimulate colony formation, other bacteria like E. coli, common in the human gut, cannot.

Working with Jon Clardy of Harvard Medical School, a natural products chemist, the two labs identified a molecule -- a fatty acid combined with a lipid that they called RIF-1 -- that sits on the surface of bacteria and is the colony development cue produced by the bacteria.

"This molecule may be betraying the presence of bacteria," Alegado said. "Bacteria just sit around blebbing off little membrane bubbles, and if one of them has this molecule, the choanoflagellates all of a sudden say, 'Aha, there are some bacteria around here.'"

The signal sets off a predetermined program in the choanoflagellate that leads to cell division and the development of rosettes, she said. The molecule RIF-1 is remarkably potent; choanos detect and respond to it at densities that are about one billionth that of the lowest concentration of sugar that humans can taste in water.

"We are investigating this molecule from many sides. How and why do bacteria make it? How do choanoflagellates respond to it, and why?" King said. She and her team also are analyzing the genome of the colony-forming choanoflagellate and the colony-inducing bacteria for clues to their interaction.

King hopes that this unexpected signaling between choanoflagellates and bacteria can yield insights into other ways in which bacteria influence biology, particularly the biology of the gut.

Coauthors with King, Alegado and Clardy are Zuzow, now a graduate student at Stanford University; Laura Brown, now a faculty member at Indiana University; Shugeng Cao and Renee Dermenjian of Harvard Medical School; and Stephen Fairclough of UC Berkeley. Dermenjian is now at Merck.

Tropical Depression 18 (TD18) formed over the southwestern Caribbean Sea at 11 a.m. EDT on Oct. 22, and NASA's TRMM satellite saw a "hot towering" thunderstorm near its center of circulation hinting that it could become a tropical storm soon. A tropical storm watch has been issued for Jamaica.

When NASA's Tropical Rainfall Measuring Mission (TRMM) satellite flew over the developing TD18 early on Oct. 22 at 0040 UTC (Oct. 21 at 8:40 p.m. EDT), the satellite measured rainfall rates within the low pressure area and measured cloud heights of the thunderstorms that make up the low.

TRMM noticed that the developing depression had a "hot towering" thunderstorm over 9.3 miles (15 kilometers) high. That same area also revealed heavy rainfall where rain was falling at 2 inches/50 mm per hour. Hot Towers are towering clouds that emit a tremendous amount of latent heat (that's why they're called "hot" towers). NASA research indicates that whenever a hot tower is spotted, a tropical cyclone will likely intensify within 6 or more hours, and the low pressure system intensified into a tropical depression.

Satellite data also showed that the depression also has a closed surface circulation and the banding of thunderstorms around the center has increased during the early morning hours on Oct. 22. These are all signs that the depression is getting more organized.

At 11 a.m. EDT, TD18 had maximum sustained winds near 30 mph (45 kph). It was located near 13.5 North latitude and 78.0 West longitude, about 320 miles (515 km) south-southwest of Kingston, Jamaica. TD18 was moving to the southwest at 5 mph 7 kph) and had a minimum central pressure of 1003 millibars.

The National Hurricane Center (NHC) noted that a tropical storm warning could be needed later in the day on Monday, Oct. 22 for Jamaica. Also, interests in eastern Cuba, Hispaniola, and the Bahamas should monitor the progress of this system. According to NHC, the depression is expected to drift westward and the turn to the north-northeast on Oct. 23 and 24. The forecast track takes TD18's center near Jamaica during the night-time hours on Oct. 23.

Tropical Depression 18 was born from the low pressure area formerly known as System 99L. The NHC expects TD18 to strengthen and become a tropical storm later on Oct. 22 or on Oct. 23. Once the depression becomes a tropical storm it would be renamed "Tropical Storm Sandy."

In addition to the new tropical depression, there's another area that forecasters are watching in the Atlantic. System 90L is producing showers and thunderstorms in the central Atlantic about 700 miles east-northeast of the Leeward Islands. During the morning hours of Oct. 22, the showers had become a little better organized, and the NHC gives the low a 50 percent chance of becoming the nineteenth tropical depression of the Atlantic hurricane season.

7 Steps to Turning Your Writing into a Thriving Online Business

make money writingGreat writers are in short supply.

And very few are willing to put in the time it takes to build an online business around their craft.

It takes hard work, but the rewards are great.

Build an online business and you can work from anywhere in the world as long as you have an internet connection.

You have the freedom to do what you want. You control your time. You get to decide what your life looks like, not someone else.

Interested?

Here are seven steps to help you make it happen.

1. Intersection

First, you have to find the intersection between what you?re good at, what you love to do, and what others are willing to pay for.

This could be teaching small businesses how to use content to grow their business.

It could mean helping dentists leverage newsletters to get more clients.

The possibilities are endless.

Whatever you do, avoid getting stuck in the planning phase. You will make tremendous progress when you take action and make mistakes.

2. Platform

Once you?ve found a viable niche, it?s time to build your platform.

Your platform will often be a blog or website.

It?s where you attract an audience of people that are interested in what you have to offer.

If you?re the dentist newsletter person, then dentists are the people you want to attract.

When you build a platform, you don?t have to figure out what people want. Once you?ve got a few hundred people, you can just ask.

The downside is that building a platform takes time. But you don?t have to wait for it to grow to start making a living as a writer.

3. Freelance Work

You can get freelance work right away.

When I was starting out, I got freelance writing gigs. I still do freelance work because it adds variety to my day.

Again, if you help dentists, you have to go out and find them. You have to hustle to get clients and show them the benefits of having a web presence.

You could even work at a discounted rate, or free, in exchange for being able to use that first dentist as a case study.

Most small business owners know that utilizing blogs, content and newsletters is what they should do, but they don?t have time.

That?s where you come in.

And while you?re getting freelance clients, you?re building your tribe.

4. Tribe

And by tribe, I mean your email list.

Have you ever heard someone say that the money is in the list?

Well, it?s true.

Your email subscribers will often be the most interested in your content, products, and services. Make sure you start building a list from the start.

It is what makes this business model work. It will make it easier to figure out what problems your audience has, and then create products around them.

5. Information Products

This naturally leads us to information products, such as ebooks and online courses.

We?re writers, so creating online courses is a breeze. And because you?ve built a list, you know what topic to cover.

Then you can sell it to your audience directly.

The key with information products is to test your ideas as quickly as possible. You don?t need a 200-page ebook.

You need to listen to your audience. Figure out what their biggest problem is, and offer a simple solution.

A short ebook is a good start. If people like it, you can expand from there.

I?ve done this with several of my products. One of them started off as a $9.95 ebook, and I?m just in the process of upgrading it into an online course.

6. Affiliate Marketing

Let?s not forget about affiliate marketing.

You can easily recommend products and services to your audience that complement what you offer.

As long as you keep it ethical, you should be fine. I only recommend products I use or have experience with.

When you look at how you can help your readers move towards their goals, everything becomes so much easier.

Then it?s no longer about your products, affiliate products, or anything else. It?s about how you can help them move forward.

7. Rinse & Repeat

Once you?ve reached this stage, it comes down to constant improvement.

Look at what you?re doing right. And ask yourself questions like:

  • Where could I improve?
  • How could I get more clients?
  • How could I add more value and raise my rates?
  • What are my income goals (and how can I get there?)

You may even want to work with a business coach, like I do, to constantly keep you on your toes and pushing your boundaries.

Summary

From there it comes down to how willing you are to face your inner demons.

It?s not easy to learn how to sell, or get clients, but like any skill, you can learn it.

Most people aren?t willing to put in the work.

They aren?t willing to build a platform or get freelance clients.

You don?t have to do everything right this very moment. You can start small. You can get your first client this week.

But whatever you do, do something.

Start somewhere.

That?s how you?ll learn what works and what doesn?t.

Astro surprises with weak debut after $1.5 billion Malaysia IPO

KUALA LUMPUR (Reuters) - Pay-TV firm Astro Malaysia Holdings Bhd closed flat in a weak market debut Friday on concerns its valuation was too high, underperforming other recent big listings as Kuala Lumpur's booming IPO market looked set to lose steam next year.

Malaysia has defied the market gloom that has seen the value of new listings drop by more than half in the Asia-Pacific excluding Japan this year, becoming the top IPO destination in the region on the back of several government privatizations and a strengthening economy.

But after such a robust 2012, and with Astro's $1.5 billion sale marking the last major listing until the first quarter of next year, the market is expected to cool off.

"For Malaysia, we will see some setback because all the bigger ones have been listed this year," said Kaladher Govindan, head of research at TA Securities.

"Most of them are somehow government-linked companies. So you don't have bigger private entities getting listed. It may run out of steam in the second half of next year."

Astro shares rose as much as 3.7 percent before erasing gains to close at 3.00 ringgit, unchanged from the offer price in Malaysia's third-biggest IPO this year. Analysts polled by Reuters had expected a rise of at least 6 percent.

The IPO by Astro, controlled by Malaysia's second-richest man Ananda Krishnan, followed Felda Global Ventures Holdings Bhd's $3.3 billion offering in June and IHH Healthcare Bhd's $2.1 billion flotation in July.

By comparison, Felda, a palm oil firm, rose 16.5 percent higher on its first trading day. Hospital operator IHH had gained 10.5 percent on its debut.

"When compared to Felda and IHH, they are big government-linked companies, and investors are more confident in government-backed firms," said Choo Swee Kee, who oversees some 700 million ringgit worth of assets as chief investment officer at Kuala Lumpur-based TA Investment Management Bhd.

"On the other hand, Astro is more of a privately owned company, that explains its weaker share price performance on its debut today."

'UPSIDE POTENTIAL'

Astro, which also counts state investor Khazanah Nasional Bhd as a major shareholder, returned to public markets after it was taken private in 2010.

Friday's closing price gives Astro a market value of 15.6 billion ringgit ($5.1 billion), nearly double the 8.3 billion ringgit it was worth when it was taken private.

The price of 3.00 ringgit would translate to a price-to-earnings ratio of 32 times based on estimated earnings per share in fiscal 2013, TA Securities said.

"There were a lot of concerns earlier that the IPO was priced at a hefty price tag, in terms of price-to-earnings ratio," TA Securities' Kaladher said of Astro's share price performance on Friday.

Still, Astro has a near-monopoly in Malaysia's residential pay-TV market with a subscriber base of 3.1 million, which some analysts said would support the share price in the longer term.

"While its IPO valuation may not appear cheap initially, there is upside potential given the existing low pay-TV penetration of 46 percent," Kong Heng Siong and Chan Jit Hoong, analysts at OSK Research in Kuala Lumpur, wrote in a recent report.

Astro will also likely see an increase in average revenue per user as subscribers migrate to high definition TV platforms, while high entry barriers to the industry due to capital expenditure requirements would limit competition, they added.

In its IPO, Astro sold shares at the top end of a marketing range, bolstered by strong demand from cornerstone investors such as U.S. hedge fund Och-Ziff Capital Management and Standard Pacific Capital. The institutional portion of the IPO, or 20.8 percent of the total, was more than 30 times oversubscribed.

IPO PIPELINE

The flurry of deals has more than quadrupled Malaysia's 2012 IPO tally to about $7.5 billion, accounting for nearly one-quarter of all new listings in the Asia-Pacific excluding Japan this year. That compares with Hong Kong's $1.83 billion and Singapore's $3.97 billion so far this year, Thomson Reuters data shows.

In Singapore on Friday, units of Religare Health Trust, which owns hospital-related assets, plunged as much as 10 percent below the initial offering price of S$0.90 in their market debut. The trust, whose assets are managed by Indian hospital group Fortis, raised $416 million through an IPO.

Malaysia's next major listing will be the planned $1 billion offering for independent power producer Malakoff, 51 percent-owned by MMC Corp Bhd, in the first quarter of next year.

Westports Malaysia Sdn Bhd, operator of the country's busiest port, is looking to raise as much as $500 million in an IPO in the second quarter of 2013.

The founders of Malaysia's AirAsia Bhd, Tony Fernandes and Kamarudin Meranun, are also set to kick off an IPO spree in 2013 with three listings worth more than $500 million.

Astro's IPO was being handled by CIMB Group Holdings Bhd, Malayan Banking Bhd and RHB Capital Bhd. Several foreign banks were also advisers, including UBS AG, Credit Suisse Group AG, Goldman Sachs Group Inc and JPMorgan Chase & Co.

NASA's Curiosity rover is rooting around what scientists call the Martian "Promised Land"?? a place where three geological formations come together to provide a deliciously complex picture of Mars' ancient past.

Although the ultimate destination for Curiosity's $2.5 billion, two-year mission is a 3-mile-high mountain called Aeolis Mons or Mount Sharp, the rover is going to be spending the next few weeks nosing around its current site, which is called Glenelg. That name comes from a geological formation in Canada's Northwest Territories, but it's also a fitting name because it's spelled the same forward and backward. Similarly, Curiosity will be going backward and forward, retracing its steps for a while when it's time to head for the mountain.

Considering that Curiosity will be in the Promised Land for several weeks, we might as well get to know the place. These pictures from Ken Kremer and Marco Di Lorenzo show you the rover's surroundings. Kremer is a New Jersey-based journalist, research chemist and photographer; Di Lorenzo is a physicist who's a high school educator and photographer in Italy.


Both men are part of an active "amateur" community that makes use of the imagery provided by Curiosity and other Mars probes, such as NASA's Mars Reconnaissance Orbiter and Opportunity rover. "Amateur" is in quotes, because the work done by amateur image-processing gurus is such a great complement to the professional work from the Mars mission teams.

Many of these gurus hang out online at UnmannedSpaceflight.com. Some also maintain their own Mars-related websites, such as Martian Vistas, the Gale Gazette and the Road to Endeavour. I used a section of one of the Kremer/Di Lorenzo panoramas as today's "Where in the Cosmos" puzzle picture on the Cosmic Log Facebook page, and it took only a matter of seconds for Bart Salatka (and many others) to name Glenelg as the place where the picture was taken. To reward his quick wits and fast fingers, I'm sending Salatka a pair of 3-D glasses that are being provided courtesy of Microsoft Research's WorldWide Telescope project. Conservation scientists look beyond greenbelts to connect wildlife sanctuaries

We live in a human-dominated world. For many of our fellow creatures, this means a fragmented world, as human conduits to friends, family, and resources sever corridors that link the natural world. Our expanding web of highways, cities, and intensive agriculture traps many animals and plants in islands and cul-de-sacs of habitat, held back by barriers of geography or architecture from reaching mates, food, and wider resources.

A team of researchers, managers, and ecological risk assessors review the current state-of-the-art in landscape connectivity planning, offering models, case studies, and advice for coping with the uncertainty inherent in dynamic, real-world conditions in the Ecological Society of America's 16th volume of Issues in Ecology.

Connectivity doesn't always mean corridors

"The shortest path is not always the best path," said author Sadie Ryan, an ecologist at the State University of New York in Syracuse. "Connectivity is not always just a straight line of greenway that you can identify from an airplane." Connections can be conduits, or more complex extensions of habitat, looking more like a web than a greenbelt. Coastal inhabitants need the depth of the reach of tides as well as the horizontal reach of coastline habitat. Birds may be able to hop from preserve to pea-patch to backyard oasis, depending on their range and flexibility.

The need to move is most obvious for migratory animals and the large animals that need big tracts of territory. Most of us are aware of large and charismatic animals like deer, bear, or coyotes. But plants, and smaller, less itinerant animals, also benefit from connections to wider spaces.

"Landscape connectivity is as diverse as the animals that live in it," said lead author Deborah Rudnick, an environmental scientist with Integral Consulting Inc, in Seattle, WA.

On the ground, managers need to address the biology of their focal species, understanding behavior, genetics, adaptation, and habitat. They have to scale up observational and experimental data to predict interactions with other wildlife and physical features of the landscape, layering on the possibility of climate changing, waterways shifting, and human life encroaching. "It's a massive amount of info to keep in your head simultaneously. I want people to step away from this review with a sense of that complexity," said Rudnick.

No perfect solutions

Opening corridors can sometimes aid the flow of invasive species and disease, as well as the species at the heart of conservation planning. All management plans involve trade-offs?whether that means obtaining the best versus the most available land, or favoring a single endangered species at the possible expense of others in its ecological community.

Some planners prefer to focus on preserving ecosystem services, rather than specific species, in an effort to preserve an ecological community more holistically. But there is no perfect solution. Unpredictable future conditions are unavoidable complications to conservation efforts, and climate change in particular could throw a ringer into the best laid plans.

"We are no longer living in a world where we can preserve perfect habitat," said Ryan.

Climate change means wildlife will move?unpredictably

Climate change, and wildlife's response to climate change, is not a linear process. We can't expect all species simply to move to colder climes, nor expect ecological communities to move as complete units, said Ryan and Rudnick. Species have independent capacities to adapt and move, decoupled from the ecological relationships of predator to prey, pollinator to flower, or grazer to grass.

"We're seeing species moving to new territory independently, remixing existing communities and shaking conservation definitions," said Rudnick. "What do we mean by quote-un-quote conserving a community? What does it mean in the face of climate change? You cannot expect a community to stay in the place that it was in the face of major changes in their environmental conditions."

Our models, and management, must adapt to conditions changing in real time. Flexibility is not generally a virtue of government regulations. The timescale of legal decision-making is generally much shorter than we would want in order to provide communities, both ecological and anthropological, time to adjust.

Achieving connections for wildlife requires forging connections with people

Corridors and connections are often in the spaces between preserves, the mixed use spaces occupied by human communities. Bridging barriers for wildlife means bridging the needs of the people living in that landscape. It means working with communities to find solutions that are practical, and possible, said the authors, not just from the perspective of science, but also residents, farmers, and industry. A first step is finding a common language. Ryan said she doesn't talk about 'ecosystem services' in rural Uganda. Those aren't Rutooro words. "We ask, "Is the park beneficial to you?"" and locals might say, "It keeps the rain." They perceive benefit from the park, but don't describe it like a scientist. The same is true of English-speaking communities.

You build your models, said Rudnick. "Then you try to put them in the real world. Community needs?that is, human community needs?add a whole layer of complexity to real life implementation."

Florida restaurant closes, Piccadilly files Chapter 11 bankruptcy ...

Piccadilly Cafeteria is a mainstay for many of its patrons. The popular Florida location has recently closed its doors due to a Chapter 11 bankruptcy filing. Reportedly, this decision was based on 'aggressive legal' maneuvers by the restaurant's lender, Atalaya Capital Management.

When a business undergoes a Chapter 11 bankruptcy, it seeks to reorganize the debt based upon a plan submitted to the bankruptcy court. If the court approves the reorganization, a business will normally have a minimum of two years, and sometimes up to five, to meet the terms of its plan. Once this occurs, the business will emerge from the bankruptcy, many times in a much stronger position financially. The closure of this Florida restaurant appears to be part of the plan to restructure Piccadilly and allow the company to continue operating.

The regional manager, Frank Frantom, states that they will do their best to place all of the displaced 13th Street location employees at other restaurant locations. Frantom also stated that a restaurant closure is the last resort, but is a necessary decision to ensure the continuity of the Piccadilly brand. Many businesses in the restaurant industry have reported that things have been getting harder over the years and that many restaurants have been coming and going not just in Florida, but all over the nation.

With the current state of the economy, businesses in Florida and all over the nation have filed Chapter 11 bankruptcy in order to rid themselves of debt and restructure their companies in order to survive and continue operating. Filing for bankruptcy may be the responsible approach to take for any business in the restaurant industry or any other industry that is struggling to pay its bills. A successful proceeding allows businesses to survive and prosper in a poor economy. Business owners considering a Chapter 11 bankruptcy may wish to consult a knowledgeable person in order to help them understand their options and begin the process.
